Identifieation and Genetic Relationship of Potato (Solanum tuberosum L.) Cultivars and Breeding Lines via Protein, Isozyme, and RAPD Analysis






Abstract
This study was carried out to establish the effective identification system for potato cultivars and breeding lines being used in breeding programs and to evaluate their genetic relationships based on similarity indices generated by soluble storage proteins, isozymes, and RAPD analysis. Twenty five potato cultivars and breeding lines were clearly identified by the analysis of 55 band patterns in all, including protein (20), isozyme (13), and RAPD (22). Cluster analysis showed that these cultivars and breeding lines could be classified into three major groups when they were analyzed by the combined cluster analysis of protein, isozyme, and RAPD. The band maMx obtained through storage protein, isozyme, and RAPD analysis can be very useful as an identification index for potato cultivars. RAPD method was, however, found to be more effective than that of protein and isozyme in terms of potato identification. On the basis of genetic relationship analysis, Korean potato cultivars and breeding lines displayed the tendency of relatively similar to the banding patterns found in USA or Japanese cultivar groups. Thus, it can be concluded that the genetic background of Korean potato cultivars and breeding lines were largely influenced by the potato cultivar groups derived from these two countries.
